However, despite the development of new therapies, the cost of developing a new drug for the treatment of a disease or disorder is very expensive. Thus, a major challenge is to develop new techniques that can be used to identify new targets for drug development.
The small peptide ligands, natural or artificial, have been used as scaffolds for the development of novel therapeutics. Peptide phage display libraries have been employed for the selection of peptide ligands that can be linked to novel protein therapeutics. This selection approach has been successfully employed in the identification of specific ligands to a variety of targets.
The selection of peptides based on their unique binding affinities to target proteins has become a powerful tool in drug discovery. Peptides are more structurally constrained than antibodies and can be designed to bind to targets with a high degree of specificity. The idea behind this approach is to use peptides as the primary binding elements to identify a secondary structure of the target or a specific protein domain involved in binding.
Discovery of peptide ligands for target proteins has been achieved using a wide variety of phage display strategies. Phage display libraries of random or constrained amino acids have been developed and used for the selection of peptides that bind to target proteins. These include libraries of natural amino acids, as well as unnatural amino acids which are used to change amino acid specificity or potency.
